It is mainly because of the fact that in the anime, Mitsunari's lord, Hideyoshi died of an illness so the third strongest warriors (first was Nobunaga who died due to an uprising), Ieyasu decided to charged at this opportunity. Mitsunari thought that Ieyasu was trying to get rid of the Toyotomi name and to destroy what dreams Hideyoshi had. The other issue was that Mitsunari's best friend, Kiyomasa, didn't believe that they could face Ieyasu. Mitsunari wanted to protect a dream, while Kiyomasa wanted to keep living. Then there was a later uprising when Mitsunari did indeed go into hiding when Kiyomasa viewed Mitsunari as a threat to what dreams Ieyasu had; there were some who were trying to save him, others joined the uprising against Mitsunari.
So Mitsunari survived the uprising and then met Ieyasu in the land of Sekigahara. Mitsunari did win, but this did involve killing Kiyomasa and he regretted it immensely.

This is why I think Mitsunari fit this role well.
