"Sempai" (or "senpai", you can spell it either way) is a Japanese honorific used to refer to someone higher up than you - like someone in a higher year in school, or someone more senior in work.
"Notice me, sempai" is a gentle poke at romantic shoujo manga, where the main character inevitably falls in love with an upperclassman, but he fails to ever realise she exists. Hence, "notice me sempai." Like most Japanese culture, it's been corrupted by the internet into a random, nearly meaningless expression.
